Juventus tore the Sampdoria defence apart with two well-worked counter-attacking moves, sealed by Federico Chiesa and Aaron Ramsey.
The opener saw all three forwards combine, Cristiano Ronaldo spreading it for Alvaro Morata to set up Chiesa at the back post.
Maya Yoshida made a stunning sliding tackle to stop Ronaldo as he was going clear on goal, while a pair of veterans went head-to-head, as Giorgio Chiellini made a decisive block on Fabio Quagliarella.
Juve only sealed it in stoppages with another Ronaldo searching pass to Juan Cuadrado, who feinted a shot before rolling it across for Ramsey.
