Harry Kane and Co were seen heading for their flight at Stansted Airport on Monday afternoon.
A video of the Spurs squad making their way out on to the runway was posted on to Tottenham's official Instagram page.
Mauricio Pochettino's side face the Serie A outfit at 5.55pm (BST) on Tuesday evening in Group B.
UEFA begin a three-season trial of having two separate kick-off times for each matchday, as in the Europa League, this week.
Two games will kick-off at the new, earlier time of 5.55pm, while the other six matches will kick-off at the slightly later time of 8pm.
Spurs are the only English club to have two early kick-offs in the group stage.
Their trip to PSV Eindhoven next month is another to have been allocated the new 5.55pm slot.
Tottenham will be looking to bounce back from their 2-1 defeat by Liverpool in the Premier League at the weekend.

Gonna say something controversial here that many will think I’m mad for doing so, but despite Lucas Moura being by far our best player this season. I personally think it’s made our team suffer as a result, we’ve changed the shape of our team with him up front with Harry and I don’t think it suits how we play as a team. I think we play our best football with Son, Dele and Eriksen playing behind Harry up top on his own. It’ll be a brave decision to drop Lucas because he has been very good, but sometimes the best players don’t make the best team and I believe that’s the case here
