The incident quickly gained attention on social media as fans were left in disbelief to see the midfielder walk off the pitch and climb over the advertising boards and into the stand.
In one video, Dier can he heard telling the fan: 'He's my brother!'
Mourinho said after the defeat that Dier had told him that he was badly insulted by the supporter and that led him to jumping into the crowd.
'I don't think that belongs to the game,' Mourinho added.
'The game was a fantastic representation of the FA Cup, but I cannot run away from the question.
'I think Eric Dier did what we as professionals cannot do but when someone insults you and your family is involved, especially your young brother.'
Mourinho went on to add that he would not agree if Tottenham chose to punish Dier over his conduct at the end of the game.
Dier is facing an FA charge for an extraordinary incident which overshadowed their FA Cup exit against the Canaries.
The England international was heard on camera footage taken by one supporter near the incident shouting 'he's my brother, he's my brother' before he was ushered away from the flashpoint and through a door at the back of the stands by bystanders, including other fans and stewards.
Dier's brother was invited into the dressing room following the clash and the club are working to identify the fan - who ran off - that was singled out for abuse by Dier.

All these Tottenham players are a disappointment. They play the beautiful game like they left their senses at home.

Someone was giving Eric Dier stick. Dier jumped in the stand, the bloke ran away and Dier literally chased him up to row 20 and down the gangway. Never seen anything like it.
