Marcelo had rifled Madrid into the lead on 16 minutes, rounding off a superb team-move that stemmed from a new look three-man defensive system.
But Diego Perotti then smashed home the leveller at the far post for Roma. The hosts were promptly pegged back, however, when Casemiro finished smartly beyond Pau Lopez to restore the lead for Zinedine Zidane's side.
Edin Dzeko rounded off the scoring in Italy, after the towering front man guided a trademark header past the rooted Thibaut Courtois.
Madrid were then downed on penalties, after Marcelo squandered the chance to restore parity from 12 yards.
