Ange Postecoglou is eager to finalise a deal for the 26-year-old.
Talks for another goalkeeper, £40m-rated David Raya of Brentford, continue.
Hugo Lloris, who has been at Tottenham since 2012, is set to leave the club this summer.
Tottenham have also asked about Bayer Leverkusen defender Edmond Tapsoba.
Vicario is part of the Italy squad but has yet to be capped by his country.
He impressed for Empoli last season and previously played for the club while on loan from Cagliari.
Vicario, who also had a loan spell at Perugia, played for Venezia between 2015 and 2019.
Burkina Faso international Tapsoba is a player that Tottenham have admired for some time.
The 24-year-old is eager to join Tottenham and it seems likely that a deal will happen once an official approach is made.
